Single molecule spectroscopy has revolutionized numerous scientific disciplines, spanning fromcatalysis, plasmonics, and polymer physics to biophysics, cell biology, and DNA sequencing. By enabling thedetection and characterization of individual molecules, this techniaue has provided unprecedented insight.into molecular interactions and dynamic processes. Moreover, single molecule approaches have played apivotal role in pushing the resolution of fluorescence microscopy beyond the difraction limit, leading tosignificant advancements in imaging.In this presentation, l will provide an overyiew of recent single molecule experiments conducted in mylaboratory, highlighting how these studies have been closely intertwined with the development of novelmicroscopy modalities, the synthesis of specialized fluorescent labels, and advances in linker chemistry. Akey focus will be our development of FLUOROcODE, a super-resolution optical mapping approach for DNA.which enables precise ysualization and structural analvsis of penomic seauences at the nanoscale. l wiillustrate how fundamental research in this domain has transitioned into real-world applications, includingmicrobiome analysis, and discuss ongoing valorization efforts through Perseus Biomics. This workunderscores the synergy between cutting-edge microscopy, innovative chemistry, and appliedbiotechnology, paving the way for new diagnostic and analytical tools in the life sciences.


主讲人简介:
JohanHofkens教授在鲁汶大学(KULeuven)获得化学硕士和博士学位,分别在大阪大学Masuhara教授和明尼阿波利斯大学Barbara教授课题组进行博十后研究。随后,他重新加入了比利时鲁汶大学,在DeSchrvver教授团队负责单分子单元项目。2005年,他被任命为鲁汶大学研究教授,2008年,晋升为正教授。2020年成为MPIMainz研究员并与浙江大学和江南大学开展合作。主要研究方向为快速光谱学、单分子光谱学和荧光显微镜,以及新型显微镜模式的开发。他致力于在生物物理学和(纳米)材料研究中应用定制技术。
